An overview of pyrazolo[3,4-d]pyrimidine heterocycles: recent synthetic approaches and biological activities

Abstract:
This review provides an up to date perspective on recent progress in synthetic strategies for constructing pyrazolo[3,4-d]pyrimidine frameworks, compounds of significant importance in both synthetic and pharmaceutical chemistry. It examines various efficient synthetic routes for creating diverse and functionalized pyrazolo[3,4-d]pyrimidine scaffolds using innovative methods. These construction methods utilize numerous approaches based on different starting materials, including those derived from pyrazole and pyrimidine, as well as miscellaneous methods such as reactions including acyclic intermediates, one-pot synthesis, solid-phase synthesis, and Diels-Alder reactions. Furthermore, the review highlights the pharmacological significance of pyrazolo[3,4-d]pyrimidines, citing a variety of activities including antimicrobial, anticancer, anti-inflammatory, xanthine oxidase inhibitors, antiviral and antihypertensive effects. This review is intended to support future researchers in the development of bioactive structures based on this versatile scaffold.
