CirculatingmiRNAsgatheredmuchinterestincancerresearch
asnoninvasivebiomarkers.Theaimofthisstudywasto
analyzetheexpressionofmiR-29candmiR-149among
colorectalcancer(CRC)patientsandtoexploretheir
diagnosticandprognosticpotentialsinrelationtotheclinical
andpathologicalfeatures.TheexpressionlevelsofmiR-29c
andmiR-149wereevaluatedintheseraof80CRCpatients,80
colorectaladenoma(CRA)patients,and80healthycontrols
usingquantitativerealtimepolymerasechainreaction(PCR).
Carcinoembryonicantigenserumlevelswereassayedusing
enzyme-linkedimmunosorbentassay.miR-29candmiR-149
weresignificantlydownregulatedamongCRCpatients
comparedwithCRAandcontrols(miR-29c,0.54±0.19vs.
0.86±0.12,0.99±0.07,P<0.001,respectively;miR-149,
0.46±0.19vs.0.74±0.012,1.0±0.22,P<0.001,respectively).
miR-29candmiR-149significantlyassociatedwithadvanced
stagesofCRC,tumorsize,andlymphaticmetastasis.Byusing
receiveroperatingcharacteristiccurveanalysis,combined
miR-29candmiR-149revealedthehighestdiagnosticpotential
forCRA(areaunderthecurve[AUC]=0.967)fromhealthy
controlsaswellasthediagnosisofCRC(AUC=0.98)from
CRA.Moreover,combinedmiRNAsrevealedhighdiagnostic
potentialfortheearlierstagesofCRCcomparedwith
advancedstages(AUC=0.96).Inconclusion,combinedserum
miR-29candmiR-149expressionanalysisestablishednovel
noninvasivebiomarkerforearlyCRCdiagnosis. ©2020
